Methods Cell Culture-The GH-cells were grown as monolayers in Ham's F-10 medium supplemented with 2.5% fetal calf serum and 15% horse serum F-10 + ; at 37 "Cunder 5% COZ, 95% air, as previously described 20 ; . In general, sets of forty-eight 35-mm dishes were used for each secretion experiment and these dishes were inoculated with approximately 2 X lo5 cells and used when the cultures reached a density of 1-2 X lo6 cells per dish. In all cases, the growth medium waschanged 24 to 48 prior to the experiment. Normal pituitary cells were obtained by enzymatic dispersion of pituitaries from lactating Sprague-Dawley rats using the method described by Ben-jonathan et al. 21 ; . If the medium samples were not assayed on the day of the experiment, they were immediately frozen a t -20 "C for later assay. The dishes were rinsed three times in cold phosphate-buffered saline, and the cells were solubilized in 1 mlof 0.1 N NaOH for protein determination by the method of Lowry et al. 22 ; using bovine serum albumin as thestandard. For C1- exchange experiments, Hanks' balanced salt solution HBSS ; was prepared according to the Gibco formulation, except that 10 m HEPES was added in all cases pH 7.4 ; , and a variable M fraction of the sodium and potassium chloride was replaced with sodium or potassium isethionate. Osmolality was kept constant at 300 mosm. Cells were rinsed once in the appropriately substituted salt solution, the medium was replaced with the same solution, and the cells were allowed to equilibrate for 15 min a t 37 "C. The dishes were then rinsed twice in HBSS with the appropriate salt, and the medium was replaced with 1 ml of the same solution with or without secretagogue. The dishes were incubated for 20 min at 37 "C 95% air, 5% COz as described above, and prolactin secretion was determined. Prolactin was measured by specific radioimmunoassay using reagents and methods supplied by the Hormone Distribution Program of the National Institutes of Health. Figures and tables show the mean and S.E. of results obtained from triplicate dishes. The statistical significance of differences among groups was evaluated using Student's t test. The binding of [3H]methyl-TRHto intact GH tumor cells was measured as previously described 23 ; . Cell Culture in Exchange Media-For experiments involving growth of cells in media with chloride replaced with isethionate, medium was made according t.o the formulation given in the Gibco catalogue except that chloride salts were replaced with isethionate.
