A relative detergent concentration of 0.3 removed the lipid Fig. 7a ; but the cores remained RNase-resistant Fig. 7 b ; . threefold higher concentration of detergent was required before any sensitivity to R N was seen, suggesting that this much was required to modify the structure of the core particle. In parallel experiments, m R N A - transcriptase activity and the protein composition of the cores were determined. Transcriptase activity of cores was measured without adding more detergent or with the addition of 0-25 ~o NP40 which gives a m a value. As the quantity of detergent used to prepare cores was increased the transcriptase activity of the cores in the absence of any free detergent also.

Rahi and colleagues report that, "distinguishing, at a population level, between the lives of people with amblyopia and those without in terms of important educational, health, and social outcomes may be difficult."1 Understanding of amblyopia has moved beyond the traditional concept of a "lazy eye" to the knowledge that it is a form of cerebral visual impairment, caused by a disturbance of vision during a sensitive period of development. Amblyopia is the effect on the developing visual system of another pathology--often refractive error or strabismus squint ; --and is the most common cause of reduced visual acuity in one eye ; in children and young adults, with a generally accepted prevalence of 2-3%.2 Clinical and experimental data, indicating better results from early treatment of amblyopia, have led to the development of childhood visual screening programmes, which detect around 7% of children as abnormal, usually because of reduced visual acuity or strabismus. Reduced visual acuity detected at screening may be due to refractive error only, in which vision immediately corrects to normal with glasses, or to amblyopia, in which a residual visual deficit exists even with refractive correction. Rarely, other pathology such as congenital cataract or retinoblastoma may be discovered. Associations between performance at school and amblyopia are complicated by the independent associations of strabismus and refractive error with a variety of neurodevelopmental disorders, including those caused by premature birth. Nevertheless, bilateral visual deficits which were excluded from Rahi and colleagues' study ; that cannot be corrected with glasses are clearly associated with educational difficulty and reduced life chances.3 Although bilateral refractive errors are relatively common in children, bilateral amblyopia is rare and a and enbrel.
Compare these two reports directly because of incomparable experimental conditions and enzyme sources; however, these results strongly indicate that the inhibitory effect of organic solvents on CYP3A4 activity is dependent on the probe drugs and or substrates. To evaluate CYP enzyme activity in vitro , the Food and Drug Administration of the US recommends the use of at least two different kinds of specific substrates. Midazolam is the second most commonly used probe drug for evaluation of CYP3A enzyme activity in vitro, reported by the US FDA. In midazolam metabolism, however, a modulatory effect of organic solvents is not clear. Studies evaluating the inhibitory potency of chemicals frequently use organic solvents to dissolve the water insoluble chemicals, resulting in assessment of the inhibitory potency by comparison with the organic solvent containing incubation media, as the `active control'. In a recent study to evaluate the drug metabolism pathway and or drug drug interaction, several organic solvents were used as dissolving solvents of the substrate or inhibitor, despite the concern of its inhibitory effect on drug metabolising enzymes. For these reasons, the present study was designed to 1 ; evaluate the effects of commonly used hydrophilic organic solvents on CYP3A activity; and 2 ; evaluate the modulation effect of the solvents on CYP3A inhibition by ketoconazole, a chemical inhibitor of CYP3A activity.
