The GTP microtubules used in this study are assumed to contain mostly GDPtubulin throughout the polymer and GTP-tubulin only in a monolayer cap at the plus end Tran et al., 1997 ; . Li and co-workers report two equilibrium dissociation constants for taxol binding to GTP microtubules: KD 61 6 and KD 3: mM: The higher affinity constant is associated with the GTP-tubulin and the lower affinity constant with the GDP-tubulin in the polymer. The KD for GDP microtubules is KD 2: mM; which is similar to the low affinity constant in GTP microtubules Li et al., 2000 ; . Throughout this article, we assume that taxol and botax bind to GTP microtubules with the same affinity, KD 3: 6 mM: GTP microtubules were observed with four different final concentrations of taxol: 93 nM, 1.3 mM, 13 mM, and 55 mM, corresponding to 2.7%, 28%, 80%, and 94% filled binding sites on the microtubules, respectively. The range of taxol concentrations was limited by the instability of GTP microtubules at low concentration and the insolubility of taxol at high concentration. Samples of GTP microtubules were made as follows: 5 ml of mg ml tubulin in GTP-PEM with 10% glycerol and no taxol was incubated at 378C for 20 min to polymerize microtubules. After polymerization, the sample equilibrated in botax for 1520 min at room temperature. The first equilibration of the highest taxol concentration 55 mM ; was with a 15: 85 mixture of botax: taxol instead of pure botax to limit the DMSO to %. Over 12% DMSO has been shown to form sheets instead of microtubules Robinson and Engelborghs, 1982 ; . The microtubules were pelleted by centrifugation at 14, 000 3 g for 15 min at 378C. The supernatant was discarded, and the pellet, which was pink due to botax on the microtubules, was resuspended in 10 ml GTP-PEM-dex with botax at the desired final concentration. The sample was equilibrated for 1 h at room temperature before being inserted into the flow cell. All manipulations were performed in the dark so as not to bleach the botax fluorophore, and samples were used within two days of preparation. Because GTP microtubules have a relatively high critical concentration even in the presence of taxol ; 8 mM; Derry et al., 1995 ; and undergo dynamic instability, the amount of tubulin polymerized into microtubules and the amount of taxol in solution are co-dependent. Upon resuspending the pellet in buffer without tubulin, microtubules will disassemble, releasing taxol into solution to maintain the critical concentration of tubulin in solution. As a result, the final taxol concentration in solution and, consequently, the fill ratio on the microtubules increase. POLYMORPHISM Unlike deliquescence, polymorphism is a very common problem in the pharmaceutical industry and huge sums of money are spent trying to identify polymorphs and trying to make stable, suitably soluble forms. Failure to resolve such problems can result in significant stability problems for the final dosage form. Ion exchange resins present a unique way to deal with the problem. A drug resinate is an amorphous solid that cannot crystallize or even form hydrates. In addition the release of the drug from the resinate is independent of the crystal form that was used to make it. Consequently, using resinates completely eliminates any problems with polymorphism.

Objective: We conducted a phase I trial of escalating doses of weekly paclitaxel Taxol ; in combination with a fixed systemic administration of 5-fluorouracil 5-FU ; in patients with advanced or metastatic gastric cancer. Methods: Patients with advanced or recurrent gastric cancer were treated with escalating doses of weekly paclitaxel as a 60 min intravenous i.v. ; infusion, along with a fixed dose of continuous 5-FU infused over 5 days. Plasma sampling was performed to characterize the pharmacokinetics and pharmacodynamics of paclitaxel. Results: Eighteen patients received combination therapy at four dose levels of weekly Taxol, ranging from 60 to 90 mg m2 week. Dose-limiting toxicities grade 3 were observed at the 90 mg m2 week dose level. Toxicities included anemia, neutropenia, thrombocytopenia, nausea and alopecia. Two episodes of grade 4 neutropenia occurred in two of the three patients receiving this dose. At each dose level, pharmacological studies documented the persistence of significant serum paclitaxel levels over 24 h after drug administration. The maximum tolerated dose MTD ; for this regimen was 90 mg m2 week of paclitaxel for 3 weeks plus 600 mg m2 day of continuous 5-FU for 5 days. Conclusions: The combination of weekly paclitaxel and 5-FU demonstrated an acceptable toxicity profile and feasible pharmacokinetic results suggesting its practical applicability. Based on these findings, the recommended dose and schedule for phase II study of combination chemotherapy is paclitaxel 80 mg m2 week 3 over 4 weeks, and continuous 5-FU 600 mg m2 day 5 days every 4 weeks. The screen would not block the light but would simply filter it so that some of the incoming infrared radiation cannot reach the Earth's atmosphere. The ones who support this scheme claim that it would cost nothing to operate and people living in the Earth can never see the mirror, so that our daily lives would not be affected. It was five o'clock in the morning and freezing. Cars filed into the dark lot at the Catholic Charities building in Des Plaines as trunks were emptied of winter coats and scarves and hats. Dozens of us gathered for pre-dawn training and a pep talk prior to searching the suburbs for people who were unsheltered and homeless. Why did we do this? To count them. Congress and the U.S. Department of Housing and Urban Development believe that understanding the size and characteristics of the entire homeless population in a community not just people using shelters is essential to the effective planning and provision of assistance and prevention services. Locally, the Alliance to End Homelessness in Suburban Cook County, on whose board WilPower sits, is the agent that coordinated the count for suburban Cook County. From Chicago Heights to Arlington Heights, pairs of volunteers were trained in how to approach a person sleeping or staying in a "place not meant for human habitation." The Alliance will use the information collected by these volunteers to learn and teniposide.

Summary We have investigated the ability of estramustine to bind to rat brain microtubule-associated proteins MAPs ; and purified MAP-2 in vitro. [3H]estramustine's relative affinity for tubulin and MAPs was assessed by gel filtration chromatography, immunoprecipitation and binding assays. Scatchard analysis demonstrated a specific affinity of the drug for MAP-2. Calculations from kinetic parameters and non-linear regression analysis gave a Kd of JM, and a Bmax of 3-4xlO~7Mml"'. Extrapolation of this value suggested that each MAP-2 molecule binds approximately 20 molecules of estramustine. Microtubule assembly studies and SDS-polyacrylamide gel electrophoresis revealed that at 20-60 uM levels, estramustine inhibited the association of MAPs with taxol microtubules. Turbidity 350 ; studies further demonstrated that 20-60 .IMestramustine inhibited MAP-2-driven tubulin assembly and produced microtubule disassembly. Electron-microscopic studies confirmed the centrifugation and turbidity results. The data demonstrated that estramustine can bind MAPs and MAP-2 specifically, thereby inhibiting microtubule assembly and taxol.
